(MALDI-TOF) Mass Spectrometry

Our lab is equipped with a Micromass TofSpec-2E MALDI-TOF instrument with delayed extraction and ion reflector. This instrument is capable of analyzing subpicomole amounts of peptides of mass less than 6 kDa with a mass accuracy of 50 PPM, and approximately 100 PPM for proteins up to 20 kDa using internal calibration. Analysis of larger proteins requires more material (in the 1-10 picomole range), and the resolution and mass accuracy of the instrument decline rapidly at higher masses. Mixtures of proteins and peptides may be analyzed, and salts and some detergents are fairly well tolerated. Samples should be in a volume of approximately 1-5 ul.
